摘要
笔者通过比较不同成分脱囊液的子孢子脱囊效果和不同蔗糖密度梯度分离纯化堆型艾美耳球虫子孢子的纯化效果来探索较佳的分离纯化堆型艾美耳球虫子孢子的条件。通过试验证明10%鸡胆汁+0.75%(W/V)胰酶脱囊效果最好。75%(W/V)和20%(W/V)蔗糖密度梯度离心得到的子孢子回收率最高,且纯度最好,染色证明纯化后的堆型艾美耳球虫子孢子仍具有较高的活性。
Through the intercomparison of excystation fluid with different components and different sucrose density gradients separation,optimized condition of Eimeria acervulina sporozoites purification was investigated. The study showed that 10% chicken bile and 0.75% （w/v）solution of trypsin had the best result 75% and 20% sucrose density gradients centrifugation obtained the best recovery rates and the purity of sporozoites ,and the result of dying showed the activity of the sporozoties is very high.
