摘要
通过紫外和荧光光谱法研究了吐温与牛胰蛋白酶作用的性质,用滴定法研究了吐温对牛胰蛋白酶活性的影响,研究表明:吐温与牛胰蛋白酶的相互作用能改变芳香族氨基酸残基在空间结构中所处的微环境,使色氨酸残基所处环境的疏水性增强;吐温对牛牛胰蛋白酶荧光猝灭效应是静态猝灭,牛胰蛋白酶内部的酪氨酸和色氨酸残基与吐温发生的能量转移作用较小,主要是牛胰蛋白酶与吐温形成复合物所引起的;吐温与牛胰蛋白酶的主要表现为疏水作用力;吐温对牛胰蛋白酶活性的影响较少。
The character of interaction between tween and bovine trypsin was studied by ultraviolet spectra and spectrofluorometric method , and the tryptic activity also was investigated by titration . The result shows that interaction of tween and bovine trypsin can change microenvironment of spatial structure of aromatic amino acid and bring the enhance of hydrophobicity of microenvironment of tryptophane ; the act of fluorescence quenching of tween and bovine trypsin is static quenching. The interaction of the energy transfer between the tyrosine and tryptophane residue of the bovine trypsin inside and tween is less and the formation of compound is principal factor. There is major hydrophobic interaction between tween and bovine trypsin. The activity of bovine trypsin is less influenced by tween.
