摘要
目的:探讨血管内皮生长因子165(VEGF165)基因对胃癌细胞侵袭性的影响及可能机制。方法:通过体外培养人胃腺癌细胞BGC-823,观察胃癌细胞转染VEGF165基因对其迁移能力及侵袭相关分子MMP-2、u-PA mRNA表达的影响。应用Millicell小室分析VEGF165转染前后胃癌细胞迁移能力的变化;逆转录聚合酶链反应(RT-PCR)研究VEGF165转染前后BGC-823的MMP-2、u-PA mRNA表达变化。结果:迁移实验结果显示Ad-VEGF165组胃癌细胞迁移能力高于Ad-GFP组及对照组[(212.000 0±10.148 8)vs(142.666 7±14.502 8)and(148.333 3±7.571 8),P<0.05];RT-PCR结果显示VEGF165转染后促进了BGC-823的MMP-2、u-PAmRNA表达,Ad-VEGF165组MMP-2、u-PA的mRNA水平明显高于Ad-GFP组及对照组[MMP-2 mRNA:(0.664 6±0.048 6)vs(0.409 6±0.066 8)and(0.380 3±0.025 4);u-PA mRNA:(0.821 6±0.079 8)vs(0.406 0±0.115 8)and(0.404 3±0.062 0);均为P<0.05]。结论:经VEGF165转染后,胃癌细胞迁移能力增强,MMP-2、u-PA的mRNA表达增加,提示VEGF在肿瘤的侵袭过程中发挥促进作用,MMP-2、u-PA可能与此作用有关。
Objective To study the effects of vascular endothelial growth factor 165 ( VEGF165 ) on the invasion of human gastric cancer cells and it' s underlying mechanism. Methods Human gastric cancer cell line BGC-823 was used in this study. BGC-823 was transfected with Ad-GFP and Ad-VEGF165. A Millieell cell culture chamber assay in vitro was used to detect the ability of migration of BGC-823. The influence of VEGF165 on the mRNA expression of matrix metalloproteinases-2 (MMP-2 ) and urokinase-type plasminogen activator (u-PA) was investigated by reverse transcription polymerase chain reaction (RT-PCR). Results Migration assay showed that the migration rate of BGC-823 was significantly higher in the Ad-VEGF165 group than that in the Ad-GFP and control groups[ (212.000 0 ±10. 148 8) vs (142. 666 7± 14. 502 8 ) and ( 148. 333 3± 7. 571 8 ), P 〈 0.05 ]. RT-PCR displayed that the VEGF165- transfected BGC-823 increased the expression of MMP-2 and u-PA mRNA,which was higher in the Ad-VEGF165 group than in the Ad-GFP group and control group [ MMP-2 mRNA: (0. 664 6 ±0. 048 6) vs (0. 409 6 ±0. 066 8) and (0. 380 3 ± 0.025 4);u-PAmRNA:(0.821 6 ±0.079 8) vs (0.406 0±0.115 8) and (0.404 3 ±0.062 0);P〈0.05]. Conclusions VEGF165-transfected can enhance the migration ability of human gastric cancer cell BGC-823, the expression of MMP-2 and u- PA mRNA was higher in the Ad-VEGF165 group than in the Ad-GFP group and control group. Which prompt VEGF in the invasion of tumor plays a catalytic role. This effect may be related to the up-regulated expression of MMP-2 and u-PA.
