摘要
为了建立有效的毛白杨二次遗传转化体系,优化了培养基激素配比、抗生素筛选浓度和乙酰丁香酮浓度。采用卡那霉素和潮霉素筛选,GUS组织染色和基因组DNA的PCR检测,通过二次遗传转化将GUS基因和潮霉素磷酸转移酶基因Hpt导入到毛白杨中,获得了60.7%二次转化频率。
To establish retransformation system for Populus tomentosa, the factors which could affect genetic transformation, including hormones combination and concentration of acetosyringone were studied. Selected by kanamycin and hygromycin, transgenic plants were identified by using GUS staining and genomic DNA PCR. The GUS and Hpt genes were transformed into poplar plants through retransformation and 60.7% transformation efficiency was obtained.
