摘要
本文探讨了植物外植体消毒技术与接种污染率的关系.结果表明:在对山楂、桑树、樱桃冬芽、郁金香球茎接种消毒时,采用外值体适当脱水(含水率保持30%左右)、消毒时剥去外被鳞片或表皮、适当延长消毒时间(樱桃冬芽除外)的方法,同时配合其它消毒措施,组织培养污染率可降低至5%以下.此技术对生长分化能力无任何影响,其中外植体剥去鳞片或表皮后进行消毒是减少污染的关键.经几年来多次试验证明,此技术比较可靠.
The relationship between the sterile techniques of the explants and contamination rate in the artificial inoculation was studied. The results showed that the techniques not only could effectively reduce the contamination rate of the explants, but also had no influence on their growth and differentiation.The main points of the sterile techniques are as follows : 1. To dehydrate the explants in moderation; 2. To peel off the scales or cuticula of the explants before sterilization; 3. To prolong the sterilization time properly according to the different kinds and types of the explants .
