摘要
本文报导了单硝酸异山梨酯缓释片(Ⅰ)的制备,以及辅料羟丙甲纤维素(HPMC)、聚维酮(PVP)对该片剂质量的影响;Ⅰ的体外释放属一级动力学过程,释放速率为0.0716h^(-1);6名健康志愿者单剂量(40mg)口服Ⅰ后血药浓度-时间数据经计算机程序处理,求得体内吸收分数。
The yield of tablets obtained in preparing Isosorbide-5-Mononitrate(5-ISMN,I)sustained release tablets was affected by the viscosity, proportion and quantity of hydroxypropyl methylcellulose (HPMC)to the release rate,the humidity and pressure to the preparution process,and the additional povidone (PVP) to the internal quality. The release profile could be described by first.order kinetics with a release rate constant of 0. 0716h-1. I was administrated orally to healthy volnteers by single-dose. The blood concentration curve was demonstrated to fit an one compartment model with frist-order absorption and first - order elimination for I and the fraction absorbed was gotten.
