摘要
目的:利用动脉粥样硬化(AS)模型观察保心丸对 AS 家兔血浆内皮素及主动脉壁内皮素免疫组化的影响。方法:喂饲胆固醇、猪油造成家兔 AS 模型。保心丸防治组同时给予保心丸8g/kg,诺衡防治组给予诺衡80mg/kg,2个月时放免测定血浆内皮素含量,用免疫组化法观察主动脉壁内皮素阳性颗粒的表达。结果:模型组血浆内皮素明显升高,两防治组均明显低于模型组。模型组主动脉内膜、中膜均见到散在的含内皮素阳性颗粒的平滑肌细胞(SMC),尤以增生活跃、向内膜下迁移的 SMC 胞浆中为多。保心丸与诺衡防治组内皮素表达明显减少。结论:保心丸和诺衡的抗 AS 作用可能与抑制血管壁 SMC 合成、分泌内皮素、降低血浆内皮素含量有关。
Objective:To observe the influence of Baoxin pill (BXP) on plasma endothelin contents and aorta endothelin im- munohistochemistry in experimental atherosclerotic rabbits.Methods:Each Japanese white rabbit was fed with 1g of cholesterol and 4g of pig oil per day for two months to induce atherosclerosis.During the same period of time,rats of BXP group were fed with BXP 8g/kg body weight,and rats of the control group were fed with Gemfibrozil 80mg/kg body weight.Radioimmune and immunohistochemical methods were used to detect the plasma endothelin level and endothelin positive granules of aorta when the formation of modal finished.Results:Plasma endothelin content in normal group was obviously lower than that of hyperlipidemia group.In hyperlipidemia group,lots of endothelin positive granules were pre- sent in aortic imima and media in particular,in the cytoplasm of smooth muscle cells (SMC) which were proliferating ac- tively and migrating toward the intima.In the two groups treated with drugs,endothelin positive granules decreased be- cause of the reduced proliferation of SMC.Conclusion:The mechanisms of the anti-atherosclerosis effect of the two drugs might be related to the reduction of plasma endothelin and the reduction of endothelin from SMC.
