rhGH、IGF-1对烧伤小鼠CD14 mRNA转录的影响

摘要 目的:探讨rhGH、IGF-1对严重烧伤小鼠早期腹腔巨噬细胞(Mφ)CD14mRNA转录的影响。方法:在体实验:20只小鼠被随机地分成A、B组(烧伤前连续5d分别给予rhGH和生理盐水)和C、D组(假烫前连续5d分别给予rhGH和生理盐水)。离体实验:10只小鼠被分成E、F组(烧伤和假烫组),Mφ加入rhGH(0,1.6,8,40ng/ml)和IGF-1(0,0.2,1,5μg/ml)培养。用细胞原位杂交方法,观察了全身应用rhGH及体外培养加入rhGH、IGF-1对小鼠巨噬细胞CD14mRNA转录的影响。结果:在体实验:(1)预先肌注rhGH,可使假烫组小鼠腹腔MφCD14mRNA的转录明显增加。(2)烧伤后腹腔MφCD14mR-NA的转录也明显增加,预先应用rhGH的小鼠烧伤后不会出现MφCD14mRNA的过度转录。(3)体外培养实验:rhGH(40ng/ml)可使烧伤小鼠MφCD14mRNA转录增加,其他浓度无显著改变,IGF-1各浓度均增加CD14mRNA的转录。结论:rhGH、IGF-1可能通过增加MφCD14基因表达使Mφ易于被激活,从而有利于免疫功能的增强。rhGH的这一作用可能通过IGF-1? Objective: To study the effects of rhGH and IGF-1 on CD14 mRNA in peritoneal macrophages in severely burned rats. Methods: In vivo: Twenty mice were randomly divided into burn group(given rhGH or NS for 5 d before burned) and sham group(given rhGH or NS). In vitro: Mouse peritoneal macrophages from burn group (n=5) or sham group (n=5) were isolated and incubated with rhGH (0,1.6,8,40 ng/ml) or IGF1 (0,0.2,1,5 μg/ml). The peritoneal macrophages CD14 mRNA expression were observed by in situ hybridization analysis. Results: Significantly increased CD14 mRNA staining granules in cytoplasm were found in burn mice (NS or rhGH) and sham group(rhGH). No difference was found between macrophages from burned mice treated with rhGH and those from burned mice injected with NS. The CD14 mRNA levels in the cells from sham or burn group incubated with all doses of IGF-1 were elevated. These changes were also observed in the cells incubated with rhGH from burn group (all doses) and from sham group (40 ng/ml).Conclusion: rhGH and IGF-1 significantly enhances macrophage sensitivity possibly because of CD14 mRNA expression changes. The increase is also seen in burn mice and could not be further enhanced by rhGH. 
