水稻不育系的广亲和基因检测及籼粳型分析 被引量:1

Identification of Wide Compatability Gene S5-n and Genetic Differentiation in Indica-japonica Property of Male Sterile Lines in Rice

摘要 根据水稻广亲和基因S5-n存在136 bp缺失的特征,设计InDel标记S5136对培矮64S、广占63S等不育系进行广亲和基因鉴定。利用与栽培稻籼、粳遗传分化密切相关的34个InDel位点,对这些不育系PCR产物的电泳结果进行判读和分析,计算其籼型或粳型基因频率(InDel分子指数法)。结果表明:粤泰A具有广亲和基因S5-n,且与来源于印度的籼稻Kasalath等存在1个相同的单碱基突变;培矮64S、N111S、C815S等10个不育系具有广亲和基因S5-n;目前广泛应用于生产的不育系培矮64S、广占63S、Y58S等的籼型基因频率均在0.76~0.91。 According to the 136 bp deletion of the wide compatibility gene(S5-n) in comparison with S5 alleles in indica and japonica varieties,S5136,an InDel marker,was designed to identify S5-n gene for the male sterile lines of Pei′ai 64S and Guangzhan 63S,etc.With 34 InDel markers closely related to genetic differentiation of rice indica and japonica varieties,the male sterile lines were analyzed for their indica gene frequency(InDel molecular method) using their individual gel electrophoresis of PCR product.the results showed that:① Yuetai A possessed the wide compatibility gene S5-n,and showed a single base pair mutation,which was similar to an indica variety,Kasalath.② Ten lines,including Pei'ai 64S,N111S and C815S,had the gene S5-n.③ Pei'ai 64S,Guangzhan 63S and Y58S,which have been widely used in the present rice production,showed their indica gene frequency in a range of 0.76~0.91.
