摘要
以融合蛋白包含体形式在大肠杆菌中高效表达了人乳头瘤病毒16型(湖北株)E7基因(HPV16E7-HB)。用蛋白质印迹技术(Westernblot)对该基因表达产物HBE7融合蛋白的性质进行了鉴定。发现HBE7融合蛋白能与HPV16E7单克隆抗体结合,表明该融合蛋白至少部分保留了E7蛋白的抗原特性。因此,将该融合蛋白免疫小鼠。3wk后,在小鼠血清中既检测到了抗载体蛋白的抗体,又检测到了特异性抗HBE7抗体。实验表明:HBE7不仅能与E7抗体产生反应,具有反应原性,而且在动物免疫中产生特异性抗体。
A recombine expressive product HBE7 fusion protein was identified by Western blot method(HPV16E7 gene from a cervical cancer patient was cloned into prokaryotic expression vector).Results showed HBE7 fusion protein could react with HPV16E7 McAb,this provided that the fusion protein at least partly reserved E7 antigen properties.Animals were inoculated with HBE7 fusion protein antigen and immune serums were prepared.Specific antibodies against HBE7 protein and a vector protein could be detected.It was found that the HBE7 protein not only had reactivity to react with E7 antibody,but also possessed strong immunogenicity to induce production of E7 antibody.This is a first report about antigenicity and immunogenicity of HPV16E7 protein in China.
