摘要
【目的】观察不同剂量的三氧化二砷(As2O3)对成年大鼠生精细胞凋亡的影响。【方法】40只健康雄性SD大鼠随机分成对照组、低剂量组(0.0375g/L)、中剂量组(0.075g/L)、高剂量组(0.15g/L)4组,灌胃法连续给药16周后对各组大鼠睾丸取材进行精子头计数,并计算睾丸脏器系数、每日精子生成量(DSP)。采用甲基绿-派诺宁染色法和TUNEL法检测大鼠生精细胞凋亡情况。【结果】①中、高剂量组睾丸精子头计数和DSP均低于对照组(P<0.05);②与对照组相比,中、高剂量组生精细胞凋亡指数(AI)均显著升高(P<0.01);③生精细胞AI与DSP呈负相关(r=-0.563,P<0.01)。【结论】一定剂量As2O3可通过促进生精细胞凋亡而导致精子生成量的减少,从而产生雄性生殖毒性。
[Objective] To investigate the occurrence of speImatogenic cell apoptosis after aniso dose arsenic trioxide (As203) administration in the testis of adult male rats. [Methods] Forty healthy male Sprague-Dawley (SD) rats were randomly divided into four groups and they were administered respectively with 0 (control group), 0.0375 g/L (low dose group), 0.075 g/L (medium dose group), 0.15 g/L (high dose group) of As203 aqueous solution by intragastric administration consecutively for 16 weeks. The numbers of testicular sperm head were counted. The coefficient of testicular viscera and daily sperm production (DSP) were calculated in each group. The apoptosis of spermatogenic cells were assessed by in situ terminal deoxynucleotityl transferase mediated dTUP nick end labeling (TUNEL) technique and methyl green-pyronin staining method. [Results] (1) In the testis of As203-treated rats, the coefficients of testicular viscera, the numbers of testicular sperm head, and DSP in medium and high dose groups were lower than that of control group (P 〈 0.05) ; (2) The apoptosis index (AI) of spermatogenic cells in medium and high dose groups significantly increased compared with that in control group (P 〈 0.01 ) ; (3) There were negative correlation between the DSP and AI of spermatogenic cells in arsenic treated rats (r = -0.563, P 〈 0.01). [Conclusions] By increasing the apoptosis of spermatogenic cells and decreasing sperm production in male rats, a certain dose of As2O3 may give rise to male reproduction toxicity. The increasing apoptosis numbers of spermatogenic cells may play an important role in leading to subfertility.
