摘要
目的观察X线造影剂(RCM)对人血管平滑肌细胞的损伤及对其增殖的影响。方法(1)人血管平滑肌细胞种于24孔培养板上至长满培养孔,暴露于250mgI/ml泛影葡胺、低渗显影葡胺、优维显、伊索显及饱和甘露醇溶液15或60分钟,收集细胞并用台盼蓝染色鉴定细胞活性。(2)人血管平滑肌细胞暴露于250mgI/ml泛影葡胺、低渗显影葡胺、优维显、伊索显、饱和甘露醇溶液及10%胎牛血清(FCS)培养液15或60分钟后,用10%FCS培养液刺激细胞增殖,增殖开始前及增殖14天后作细胞计数。结果(1)暴露于250mgI/ml优维显、伊索显、饱和甘露醇溶液60分钟及250mgI/ml泛影葡胺、低渗显影葡胺15分钟后,细胞活性无改变,暴露于250mgI/ml泛影葡胺、低渗显影葡胺60分钟后分别出现16.5%±6.6%及9.2%±7.8%的死细胞。(2)250mgI/ml泛影葡胺、低渗显影葡胺及饱和甘露醇溶液抑制血管平滑肌细胞增殖,优维显及伊索显对血管平滑肌细胞增殖无影响。结论(1)高浓度离子型RCM有细胞毒性;(2)RCM对人血管平滑肌细胞的增殖无直接刺激作用;
Purpose To observe radiographic contrast media (RCM)'s cytotoxicity on human vascular smooth muscle cells (VSMC) and their effects on the growth of human VSMC in order to find out, whether any relation exists between RCM and myointimal hyperplasia after PTA. Methods (1)Human VSMC were seeded on 24 wells plates, after growing to confluency, the cells were exposed to 250 mg I/ml of diatrizoate, ioxaglate, iopromide, iotrolan and saturated mannitol solution, then the cells were collected and their viability was examined using the trypan blue exclusion test; (2) Human VSMC were exposed for 15 or 60 min to 250 mg I/ml of diatrizoate, ioxaglate, iopromide, iotrolan, saturated mannitol solution and 10% FCS medium (control), then cell growth was stimulated with 10% FCS, cell count was carried out on day 0 and day 14. Results (1) There was no significant change of VSMC viability after 60min exposure to iopromide, iotrolan, saturated mannitol solution and after 15min exposure to diatrizoate or ioxaglate. After exposure to diatrizoate or ioxaglate for 60 min, 16.5%±6.6% and 9.2%± 7.8% dead cells were found respectively( P <0.01 vs controls). (2) In VSMC growth assay, diatrizoate, ioxaglate and saturated mannitol solution reduced VSMC growth rate ( P <0.05 vs control), no significant change was observed with iopromide and iotrolan. Conclusion (1) High concentration of ionic RCM were cytotoxic in comparison to the nonionic. (2)There was no direct stimulatory effect of RCM on human VSMC growth which might contribute to restenosis after angioplasty. (3) The cytostatic effect of RCM seemed to be dependent on both osmolality and chemotoxicity.
