摘要
目的:研究川芎嗪对心肺复苏后大鼠肾脏的保护作用及其机制。方法:36只SD大鼠随机均分为假手术组、模型组和川芎嗪组,采用窒息合并冰氯化钾停跳液致大鼠心跳骤停-心肺复苏模型,复苏后24 h检测血清尿素(SU)、肌酐(Scr)、肾组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,观察肾小管上皮细胞凋亡情况。结果:与假手术组相比,模型组SU、Scr和MDA均明显升高(P<0.01),SOD降低(P<0.01),肾小管上皮凋亡细胞增多(P<0.01)。与模型组相比,川芎嗪组SU、Scr和MDA均明显降低(P<0.01),SOD升高(P<0.01),肾小管上皮凋亡细胞减少(P<0.01)。结论:川芎嗪通过减轻氧自由基损伤和抑制肾小管上皮细胞的凋亡从而保护心肺复苏后大鼠的肾功能。
Objective To investigate the protective effects and its mechanism of ligustrazine on rat kidney after cardiopulmonary resuscitation. Methods Thirty-six SD rats were randomly divided into sham group, model group and ligustrazine group. Cardiac arrest was induced with asphyxiation combined ice-cold potassium chloride. Twenty-four hours after resuscitation, the serum levels of urea and creatinine, renal SOD activities and MDA contents were detected, the apoptosis of renal tubular cells was assessed. Results Compared with sham group, the serum levels of urea and creatinine, renal MDA contents and the number of apoptotic cells were significantly increased(P 〈 0.01, respectively), the activity of renal SOD was significantly decreased in model group(P 〈 0.01 ). In hgustrazine group, the serum levels of urea and creatinine, renal MDA contents and the number of apoptotic cells were lower than that in model group(P 〈 0.01, respectively), renal SOD activity was higher than that in model group(P 〈 0.01 ). Conclusion Ligustrazine could protect renal function in rats after cardiopulmonary resuscitation, the mechanism is probably through alleviation of oxyradical injury and inhibition of tubular cell apoptosis.
