摘要
目的:研究豚鼠脑干听觉中枢及内侧膝状体(?)受体β亚单位的分布.方法:采用兔抗GABA_A受体β亚单位血清和免疫组织化学ABC法及葡萄糖氧化酶-DAB-镍成色技术.结果:在豚鼠耳蜗核、上橄榄复合体、下丘及内侧膝状体诸核团,均有GABA_A受体β亚单位阳性结构的分布.在耳蜗前腹核可见较多的GABA_A受体β亚单位阳性神经元及纤维,阳性神经元的胞体较大,多呈圆形或椭圆形,直径约30μm~50μm,阳性产物主要位于胞膜及近体树突.耳蜗背侧核阳性神经元的数量较少.上橄榄外侧核GAB_A受体β亚单位阳性神经元较多且密集,多呈小圆形,直径约16μm~25μm.下丘中央核也可见直径16μm左右的GABA_A受体β亚单位阳性的小圆形神经元,数量较少.内侧膝状体未见GABA_A受体β亚单位阳性的神经元,而阳性纤维密集.结论;在豚鼠脑干听觉中枢及内侧膝状体,有GABA_A受体β亚单位阳性神经元及纤维广泛分布,GABA在调节听觉信号传入的过程中,可能具有重要的作用.
To investigate the distribution of β subunit of GABAA receptor in the brain stem auditory centre and medial geniculate body (MGB) of guinea pigs. Methods: Rabbit serum against β subunit of GABAA receptor, immunohis-tochemical ABC method and glucose-oxidase-DAB-nickle technique were used in this study. Results: β subunit of GABAA receptor immunoreactive, productswere distributed widely in the cochlear nucleus,superior olivet complex, inferi-or colliculus and MGB. A number of β subunit of GABAA receptor positive neurons and fibers were found. In the anterior ventral cochlear nucleus, the positive neurons, round or oval in shape with a diameter about 30μm~50μm, showed the positive products located predominately in the cell membrane and proximal dendrites. A few of positive neurons could be seen in the dorsal cochlear nucleus. In the lateral superior olive, many positive neurons, small round in shape with a diameter about 16μm~25μm were observed. The small round positive neurons with a diameter about 16um and positive fibers were also found in the central nucleus of inferior colliculus, but the number was fewer. No positive neurons could be found in the MGB, but quite dense β subunit of GABAA receptor positive fibers in the MGB were observed. Conclusion: β subunit of GABAA receptor positive neurons and fibers are distributed widely in the brain stem auditory centre and MGB of guinea pig. GABA might play an important role in modulating sound messages afferent.
