摘要
目的探讨运动对脂肪细胞脂联素mRNA表达的影响,从脂肪细胞内分泌的角度研究肥胖的机制。方法断乳5天SD健康雄性幼鼠40只,体重75-85 g随机分组为普通饲料喂养组和高脂饲料喂养组,高脂饲料喂养组在3周后筛选出肥胖鼠,并随机分组为不运动对照组和运动干预组。运动大鼠进行游泳运动,运动时间第1周为30 min/d、第2周为60 min/d(运动30 min休息5 min)、第3周为90 min/d(运动30 min休息5 min)、第4周为120 min/d(运动30min休息5 min),每周运动6 d,周日休息,共4 w。实验后处死所有大鼠,取睾丸脂肪垫和腹膜后脂肪组织测试脂联素mRNA表达水平。结果肥胖大鼠运动4周后血清脂联素浓度增加了51.81%(P<0.05),睾丸脂肪垫和腹膜后脂肪组织脂联素mRNA表达水平分别增加86.36%(P<0.05)和94.74%(P<0.05)。结论运动明显增高血清脂联素水平和内脏脂肪组织脂联素mRNA的表达。
Objective To study the effects of exercise on expression of adiponeetin mRNA in adipoeyte, and the mechanism that exercise intervened in obesity to be elucidated. Methods 40 male rats whose weight was between 75 g and 85 g were weaned for five days and divided into normal diet and high fat diet groups randomly. After 3 weeks, Obese rats were choesed and divided into control group and experiment group randomly. The rats in experiment group were dealed with swimimg training for 4 weeks, exercised six days per week and rested one day. Exercise time was 30 min per day in the fist week,60 min per week in the second week,90 min per week in the third week and 120 min per week in the last week.From second week,Rats swam for 30 min and breaked 30 min. After 4 weeks, all rats were killed and the levels of adiponectin mRNA expression were measured in testis fad pad and rctroperitoneal fat pad of rat. Results As Obese rats exercised in 4 weeks, the level of serum adiponectin was increased 51.81%( P 〈0.05).and the expression of adiponectin mRNA increased 86.36% ( P 〈 0.05 ) in testis fad pad and increased 94.74% ( P 〈 0.05 ) in retroperitoneal fat pad. Conclusions The level of serum adiponectin increased significantly , and the expression of adipanectin mRNA increased obviousiy in testis fad pad and retroperitoneal fat pad of rat after swimming training.
