摘要
目的观察SD大鼠下肢缺血模型造模后不同时间点缺血组织相关蛋白表达量的变化。方法将42只SD大鼠随机分为6组,每组7只。第1组为对照组;第2~6组为模型组,将模型组大鼠左侧股总动脉结扎离断造模,分别于造模后4h、3d、1周、2周及4周5个不同时间点分离大鼠左小腿腓肠肌组织,利用Western blotting检测法检测缺血组织酪氨酸蛋白激酶受体-2(Tie-2)及血管内皮生长因子(VEGF)的蛋白表达量。结果缺血组织Tie-2表达量在组织缺血后迅速减少,至3d时Tie-2表达量持续于较低水平,然后缓慢增加,缺血后2周时表达有较大幅度增加,但仍低于正常组织,之后有小幅度减少。缺血组织VEGF表达在缺血后4h即开始增多,至3d时达到高峰,以后逐渐减少,至缺血后2周时表达最少,然后又缓慢回升,到缺血后4周基本接近正常组织VEGF表达量。结论在缺血组织VEGF表达上调的过程中,缺血组织完成了修复,肌组织中Tie-2表达量增多,内皮细胞数量得到增加,从而改善缺血组织供血。
Objective To invest proteic expression variation at different time point of ischemi,, tissue in rats model of lower limp isehemia. Method Rats were randomized divided into 6 groups, 7 rats for each group. Th, first group was control group, the other five groups were model groups. Ligation and mutilation of rats left femoral after), at 5 different time point after building models:4h, 3d , 1 w , 2w and 4w , separated rats left lower limp gastroe muscular tissues. Through western blotting method detected proteic expression of Tie-2 and VEGF in ischemic tissue. Results The expression of Tie-2 from ischemie organizations decreased rapidly postischemia, it stayed at the low level at the 3rd day and increased slowly after then. It increased sharply at the 2nd week, still less than the expression of normal tissue, and it decreased slowly after then. Proteic expression of VEGF began to increase after ischemia. It was on the top at the 3rd day. It decreased gradually after then. It met the bottom at the 2nd week. It increased slowly After the second week. It was almost the same as the proteic expression of normal tissue at the 4th week. Conclusion The proteic expression of Tie-2 was increasd, which indicated that the number of endothelial cells increased and the organization completed the repair of ischemia during the course of increasing expression of VEGF postischemia. Endothelial cells participated in the new blood vessels, so as to improve blood supply for ischemic organizations.
