吡那地尔缺血后处理对大鼠心脏的保护作用

摘要 目的研究吡那地尔缺血后处理对离体大鼠心脏缺血/再灌注损伤的影响。方法应用Langendorff离体心脏灌流技术建立大鼠全心缺血/再灌注模型和膜片钳细胞贴附式记录技术。结果①与对照组比较,缺氧/复灌组表现出明显的心脏损伤效应,复灌过程中各项心功能指标(LVDP、±LVdp/dtmax)的恢复均低于对照组(P<0.05);与缺氧/复灌组比较,经吡那地尔缺血后处理组的顿抑心肌收缩功能恢复明显提前(P<0.01),心功能(LVDP、±LVdp/dtmax)恢复增强(P<0.05);②吡那地尔在缺血/再灌注条件下提高KATP通道的电流幅度,增加通道的开放概率,各组之间比较差异有显著性(P<0.01)。结论短暂低氧方式同时经吡那地尔缺血后处理,可增强大鼠顿抑心脏对缺血/再灌注损伤的抵抗能力,表现出明显的心脏保护作用。 Aim To observe the effect of pinacidil ischemic postconditioning on rat ventricular myocytes during ischemia/reperfusion. Methods Rat acute myoeardical anoxia-reperfusion model was used. The cardiac function and eleetrophysiological properties were surveyed by isolated perfusion and patch-clamp technique, respectively. Results (a) Compared with those in the control groups, these results demonstrated that anoxia-reperfusion groups in rats could increase PSP and - dp/dtmax markedly while LVDP, + dp/dtmax and HR deereased slightly. Significant difference was found in A/R group; while pinacidil group showed the better recovery of cardiac function after ischemia/reperfusion. (b) using patch-clamp technique, significant difference was found in pinacidil group, including increasing mean open time and conductance value. Conclusion Ischemic postconditioning by Pinacidil(PCOS of KATP) exerts an cardioprotection effect on acute myocardial anoxia-reperfusion injury.
