摘要
目的:观察雷公藤多甙(tripterygiitotorum,TⅡ)及FK506滴眼液局部应用对大鼠角膜移植免疫排斥反应的影响。方法:建立大鼠角膜移植模型,随机按局部滴用药物分组成A,B,C组,用裂隙灯记录及比较各组移植排斥指数,并比较角膜排斥发生时间。分别于术后15d及30d行常规病理学检查。并通过免疫组化检查CD25,CD5+4在角膜植片基质中的表达。结果:0.3g/LTⅡ及0.5g/LFK506抑制角膜移植免疫排斥反应的效应均有效,两者相比效果无显著性差异。HE染色及免疫组化分析可见15d时各有效组角膜植片厚度基本正常,基质中仅有少量CD25,CD5+4阳性细胞表达、淋巴细胞浸润,新生血管少见。结论:TⅡ与FK506疗效近似。具有抑制高危角膜移植免疫排斥反应发生的临床应用价值。
AIM :To observe and compare the effect of the TII, FK506 ophthalmic solutions on the corneal allograft rejection.
METHODS: The corneal transplantation model on the closed colony rats were set up and then were randomly divided into A, B and C groups by the using of different eye-drops. Graft fate was assessed clinically by slit-lamp biomicroscope examination, included the corneal clarity, edema and vascularization, and the rejection time. Respectively, after 15 days and 30 days of routine histopathological examination. And inspect expression by immunohistochemistry CD25, CD54^+ in the corneal graft stroma.
RESULTS: There were no statistical different between group 0. 3g/L TII and 0. 5g/L FK506 in prolonging the corneal survival. While comparing the histopathological and immunohistochemistry findings between the groups, the results confirmed that the topical application of TII and FK506 eye-drops could significantly reduce the inflammatory cells infiltrating the graft stroma and the neovascularization, whilst reduce the production of ICAM-1 and IL-2.
CONCLUSION: The corneal allograft rejection could be significantly suppressed by the local use of T, ophthalmic solution. 0. 3g/L T, has the same effect as the 0. 5g/L FK506 in prolonging the corneal survival.
