摘要
目的:以微透析装置连接高效液相色谱(MD-HPLC)仪分析牛奶中的6种磺胺类药物,即:磺胺嘧啶、磺胺一甲基嘧啶、磺胺二甲基嘧啶、磺胺恶唑、磺胺一甲氧嘧啶、磺胺二甲氧嘧啶的含量。方法:最佳层析及微透析条件:HPLC流动相25%CH3CN、0.01mol/LNaH2PO4(pH5.0),搭配反相C-8管柱进行洗脱;以超纯水为灌注液,流速1.06μL/min,收集20min透析液,可实现6种磺胺类药物的分离、检测。结果:在最佳色谱及微透析条件下回收率:SDA105%,SME104%,SMX93%,SMZ98%,SMM98%,SMO102%;检测极限:SDA0.41μg/L,SME0.86μg/L,SMX0.09μg/L,SMZ0.45μg/L,SMM0.19μg/L,SMO0.08μg/L。结论:本方法简便,分离效果好,灵敏度高,重现性好。对市售牛奶进行磺胺类药物含量的检测效果良好。
Objective: To analyze 6 sulfonamides, namely Sulfadiazine (SDA), Sulfamerazine (SME), Sulfamethazine (SMZ), Sulfamethoxazole (SMX), Sulfamonomethoxine (SMM), Sulfadimethoxine (SMO), in milks by micro-dialysis connected with HPLC. Methods: The optimal conditions of the micro-dialysis sampling were pure water as perfusate with the flow rate of 1.06 L/min and 20 min collection time. And the optimal conditions of HPLC analysis were C8 reversed phase column with the mobile phase of 25% CH3CN, 0.01 mol/L NaH2PO4(pH 5.0). Results: By the optimal conditions, the recoveries of micro-dialysis were of SDA 105%, SME 104%, SMX 93%,SMZ 98%, SMM 98%,SMO 102%. And the detection limits of SDA 0.41 μg/L,SME 0.86 μg/L, SMX 0.09μg/L, SMZ 0.45 μg/L, SMM 0.19 μg/L respectively. 0.151, 0.130, 0.038, 0.002, 0.002 and 0.003 μg/L were found for SDA, SME, SMX, SMZ, SMM and SMO, respectively for the system of micro-dialysis connected with HPLC. Coneulsion: The method is simple, accurate, reproducible and can offer good results to examine sulfonamides in milk.
