摘要
利用指数富集配体进化技术(SELEX)可获得与目标靶具有特异性结合能力的适配体(寡核苷酸)。经过近20年的研究,适配体被证实可在科研及临床应用中部分取代抗体,是有很大发展前景的技术领域。适配体技术发展的关键在于对目标靶具有高选择性吸附能力的适配体的筛选和获得。十几年来,以提高筛选效率和效果为目标的适配体筛选技术不断改进,产生了如消减筛选、复合靶筛选、基因组筛选、毛细管筛选等新方法,推动了这一技术的发展。本文对现有适配体筛选方法进行了系统的评述。
Aptamer(oligonucleotide) with specific binding capacity for target, could be obtained by using a screening technique called systematic evolution of ligands by exponential enriehment(SELEX). With the development for about two decades, aptamers, as an alternative to antibodies, showed bright prospect in scientific research, clinical diagnosis and therapy. As the screening technique is the key to get aptamer successfully, new approaches and methods such as subtractive SELEX, complex targets SELEX, genomic SELEX, CE SELEX appeared continually to improve the screening efficiency in recent years. These methods were reviewed in this paper.
