摘要
目的观察枸杞子提取液(LBA)对RCS大鼠遗传性视网膜变性的治疗作用。方法出生后RCS大鼠24只,随机分为对照组和治疗组,每组12只。治疗组RCS大鼠出生后10d开始喂食1mg/kg的LBA,对照组正常饲养。两组大鼠分别在出生后25、35、45d取眼球,制备组织切片,应用苏木精-伊红(HE)染色和TUNEL染色观察视网膜感光细胞坏死和凋亡情况,并应用免疫组化方法检测Caspase-2的表达。结果治疗组大鼠出生后25d和35d时TUNEL染色阳性细胞与对照组相比明显减少(t=3.33~6.72,P<0.05);治疗组和对照组大鼠生后25~45d时Caspase-2在节细胞层和内核层均有表达,但在25d时治疗组与对照组相比明显减少(t=6.33,P<0.05)。结论在RCS大鼠遗传性视网膜变性的早期,LBA通过抑制细胞凋亡及Caspase-2表达对神经元起保护作用。
Objective To investigate the neuroprotective effect of the aqueous extract of dried Lycium barbarum (LBA) on hereditary retinal dystrophy of the Royal College of Surgeon (RCS) rats. Methods Twenty four postnatal RCS rats were equally randomized to control group (CG) and treatment group (TG). In TG, the rats started a diet of 1 mg/(kg · d) LBA on the day 10 after birth, and in CG, the rats were fed with normal food. Eyeballs were obtained from both groups on day 25, 35 and 45 after birth. Histological sections were prepared with HE or TUNEL staining for observation of photoreceptor cells necrosis and apoptosis. The expression of Caspase-2 protein was detected with immunohistochemistry staining. Results The numbers of cells positively stained with TUNEL in rats of TG 25 and 35 days after birth were significantly decreased compared with those of CG (t=3.33-6.72,P〈0.05) ; The expression of Caspase-2 protein was found in the ganglion cell layer and inner nuclear layer in both groups 25 and 45 days after birth, but the number of Caspase-2 positive cells in TG was significantly smaller than that of CG on day 25 (t= 6.33,P〈0.05). Conclusion In the early stage of hereditary retinal dystrophy of RCS rats, LBA reveals a neuroprotective effect through inhibiting cell apoptosis and expression of Caspase-2 protein.
