刺参黏多糖对人宫颈癌细胞凋亡的影响 被引量:8

摘要 目的探讨刺参黏多糖(Sjamp)对人宫颈癌HeLa细胞凋亡的影响。方法以0~1.6g/LSjamp分别处理HeLa细胞72h后,以Fluo-3/AM检测宫颈癌HeLa细胞内钙离子浓度的变化,并应用流式细胞仪观察Sjamp对人HeLa细胞凋亡率的影响。结果Sjamp作用后的人HeLa细胞凋亡率升高且与药物剂量呈依赖关系(F=505.71,P<0.05);Sjamp作用后的人HeLa细胞内钙离子浓度升高且与药物剂量呈依赖关系(F=247.46,P<0.05)。结论Sjamp能剂量依赖性地诱导HeLa细胞凋亡,而细胞内钙离子浓度增高可能是其诱导凋亡的机制之一。 Objective To study the effect of stichopus japonicus sekenka (Sjamp) on apoptosis of HeLa cells in human cervical cancer. Methods HeLa ceils were treated by various concentrations of Sjamp from 0 g/L to 1.6 g/L,the effect on apoptosis was observed by flow eytometry. Calcium in cells was detected by Fluo-3/AM. Results After acting by Sjamp, the apoptosis rate of HeLa cells rose (F= 505.71 ,P〈0.05), the concentration of intraeellular Ca^2+ increased, both became dose-dependent (F=247.46,P〈0.05). Conclusion Sjamp can induce the apoptosis of HeLa cells dose-dependently, the increase of intracellular Ca^2+ maybe one of the mechanisms leading to apoptosis.
