酸性微环境下单核/巨噬细胞对舌鳞癌细胞系Tca-8113杀伤作用的实验研究

摘要 目的:以舌癌细胞株Tea-8113为研究对象,探讨酸性微环境对单核/巨噬细胞对口腔癌细胞吞噬和杀伤作用的影响。方法:从健康人的外周血中分离单核细胞并通过培养将其转化为单核/巨噬细胞;分别调整培养液的pH值为酸性:pH值为6.6、6.8;常规环境:pH值为7.2,将单核/巨噬细胞和舌癌细胞株Tea-8113进行混合培养,4h后MTT法检测单核/巨噬细胞对细胞的杀伤作用。结果:在酸性微环境,单核/巨噬细胞对舌癌细胞株的杀伤作用低于常规环境下(P〈0.05)。这说明酸性微环境可以使单核/巨噬细胞的功能发生改变。结论:由于肿瘤组织内酸性微环境的作用,浸润到肿瘤组织中的单核/巨噬细胞对肿瘤细胞的吞噬和杀伤作用减弱,可能更多的参与了肿瘤的生长和转移,但如何调节单核/巨噬细胞在肿瘤组织中的功能及在肿瘤免疫治疗中的意义还需进一步的研究。 Objective: To investigate the anti-tumor effect of monocytes/macrophages on Tca8113 cells in an acidic microenvironment in vitro. Methods: Peripheral blood mononuclear cells were extracted from healthy people's blood and treated to enrich for monocytes/macrophages. The monocytes/macrophages were cultured with Tca8113 in an acidic (pH 6.6 and pH 6.8) microenvironment and in a normal microenvironment (pH 7.2). The anti-tumor effect of monocytes/macrophages on Tca8113 cells was examined by MTT assay. Results: The anti-tumor effect of monocytes/macrophages on Tca8113 cells in an acidic environment was decreased compared to that found in a normal environment (P〈0.05). Conclusion: Due to the acidic microenvi- ronment present inside tumors, the anti-tumor effect of monocytes/macrophages on Tca8113 cells weaken, possibly promoting tumor growth and metastasis. More research is warranted to further elucidate the anti-tumor effects of monocytes/macrophages.
