摘要
目的:研究Cx26在肝细胞癌及正常肝组织中的表达情况。方法:免疫组化和RT-PCR检测Cx26和PCNA的表达。结果:①Cx26mRNA在正常肝组织组和肝癌组的表达水平分别为0.82±0.37和0.38±0.10;Cx26mRNA在HCC高分化组中的表达高于其在低分化组中的表达水平,分别为0.48±0.12和0.21±0.05。②Cx26在所有正常肝组织中均高表达,其在肝癌组织中的阳性率为64.71%,Cx26的表达随着肝癌分化程度的增加而增强。③在肝癌组织中,PCNA-LI在Cx26蛋白阳性表达组和阴性表达组分别为32.72%±4.21%和56.67%±7.21%,Cx26蛋白阳性者的PCNA-LI明显低于阴性者,PCNA表达水平与Cx26蛋白表达呈负相关。结论:①在肝癌组织中,Cx26的表达随着组织分化程度的降低而降低。②Cx26可作为评价肝癌预后的指标之一。
Objective: To study the expression of connexin 26 (Cx26) in hepatocellular carcinoma (HCC) and normal liver tissues. Methods: Immunohistochemistry and reverse transcription-polymerase chain reaction (RT-PCR) were employed to determine the expression of Cx26 and proliferating cell nuclear antigen (PCNA). Results: The expression of Cx26 mRNA in normal liver tissues and hepatocellular carcinoma was 0.82±0.37 and 0.38±0.10, respectively. The expression level of Cx26 mRNA in the well differentiated group (Grade Ⅰ-Ⅱ) was 0.48+0.12, higher than the 0.21±0.05 found in the poorly differentiated group (Grade Ⅲ-Ⅳ). Normal liver tissues were positive for Cx26 staining. The positive rate of Cx26 protein expression was 61.7% in the hepatocellular carcinoma samples. The expression of Cx26 protein decreased as the malignant degree increased. PCNA protein expression was 32.72±4.21% in the Cx26-positive group and 56.67±7.21% in the Cx26-negative group. The expression of PCNA protein was negatively correlated with the expression of Cx26 protein. Conclusion: The expression of Cx26 decreases as the differentiation degree of HCC increases. Cx26 can be used as a prognostic indicator for hepatocellular carcinoma.
