提高BrdU标记成年大鼠在体脑神经干细胞增殖和分化效率的研究

摘要 目的探讨合理利用BrdU(5-溴脱氧尿嘧啶核苷)单标成年在体脑神经干细胞增殖以及双标成年在体脑神经干细胞分化的策略。方法在BrdU通过腹腔注射6h后或在BrdU腹腔注射后14~21d将大鼠灌注固定,继之用邻片进行H—E染色以作定位,分别对脑组织切片进行BrdU单标或BrdU+NT(神经丝,成熟神经元的标志物)、BrdU+GFAP(胶质原纤维酸性蛋白,成熟星形胶质细胞的标记物)以及BrdU+GC(半乳糖脑苷脂,成熟少突胶质细胞的标记物)的双标免疫组织化学反应,光镜下观察和计数单标或双标的阳性细胞。结果BrdU单标阳性细胞多见于侧脑室的SVZ(室管膜下区)和SGZ(齿状回颗粒下区),而BrdU+NT阳性细胞多位于嗅球和齿状回的颗粒细胞层,BrdU+GFAP阳性细胞多见于SVZ或SGZ;BrdU+GC阳性细胞数最少,散在分布于以上四个部位内。结论在进行BrdU标记脑神经干细胞前应该制定优化组合方案,以获得最佳的单标和双标的识别效果。 Objective To investegate the strategies for improving the proliferation and differentiation of neural stem cells in the brains of adult rats. Methods BrdU-singly-labelled or BrdU-doubly-labelled immunohistochemistry was used to observe the capacities of proliferation and differentiation of neural stem cells in the subventricular zone (SVZ) of lateral ventricle and the subgranular zone (SGZ) of dentate gyms. Results BrdU- immunoreactive (BrdU-IR) and BrdU + NT ( neurofilament), BrdU + GFAP ( glial fibrilliary acidic protein) and BrdU + GC ( galactocerebroside)-IR cells were mainly distributed in the SVZ or SGZ as well as olfactory bulb and granular layer of dentate gyms, respectively. Conclusion BrdU is a reliable marker for labelling both proliferation and differentiation of adult brain neural stern cells as long as a good plan is made before the experiments are performed.
