摘要
为了观察硫代反义寡聚核苷酸对单纯疱疹病毒(HSV)毒力和小鼠实验性HSV角膜炎发病的影响,培养细胞中加入滴度为102TCID50的HSV和100、50、25、12.5μg/ml的反义寡聚核苷酸,培养3天,将对照病毒和与100、25μg/ml药物作用的病毒稀释至10-3,做空斑实验。将对照病毒和与50、12.5μg/ml药物作用的病毒稀释至10-2,种植于小鼠角膜,观察角膜上皮和实质病变。病毒对照组的空斑数为26.5个,100和25μg/ml药物组分别为7.5和6.0个,两药物组的空斑数极显著少于对照组,其空斑抑制率分别为71.7%和75.5%。小鼠角膜上皮病变,在第3和第8天,两药物组显著轻于对照组,角膜实质病变三组间无显著性差异。硫代反义寡聚核苷酸可使HSV的毒力下降,使病毒对小鼠角膜的感染力降低。
To observe the effect of phosphorothiate antisense oligonucleotide on HSV activity and mice HSV kera titis, 10 2TCID 50 virus and 100,50,25,12.5μg/ml oligonucleotide were added in cell culture plates The cells were cultured for 3 days The control virus and virus co cultured with 100,25μg/ml drugs were diluted to 10 -3 and plaque assay was done The control virus and virus co cultured with 50,12 5μg/ml drugs were diluted to 10 -2 and planted in the mice cornea The corneal epithialial and stroma lesion was observed The plaque number of the control group was 26 5 and those of 100,25μg/ml drug groups were 7 5 and 6 0 respectively The plaque number of each of drug group was very significantly less than that of the control group Their inhibitive efficiency was 77 7% and 75 5% respectively Mice corneal epithialial lesion of two drug groups was significantly less than that of the control group in the 3rd and 8th day There were no significant diferences among the three groups in corneal stroma lesion So we draw a conclusion that phosphorothiate antisense oligonucleotide may decrease HSV activity and lighten mice corneal infective degree to HSV
