摘要 以HPLC和动态浊度法分别检测血塞通注射液超滤前后有效成分及细菌内毒素的含量变化,研究了10kDa及50kDa截留分子量的超滤膜对血塞通注射液中有效成分的影响及细菌内毒素的去除效果。结果表明,两种截留分子量的超滤膜均可有效的去除药液中的细菌内毒素,但10kDa超滤膜对三七总皂苷有效成分有明显损失,50kDa超滤膜对成分保留率均在95%以上。选用截留分子量为50kDa的超滤膜即可保留有效成分,又可有效去除细菌内毒素,适用于血塞通注射液的生产。 For Xuesaitong injection, the recovery rate of effectual components and the elimination of bacterial endotoxins by uhrafihration membrane ,with cut off molecular weight (MWCO) 10 and 50 kDa,were studied in this work. The content of effectual components and bacterial endotoxins were detected by HPLC and kenitic-turbidimetry respectively. The experimental results showed that uhrafihration membrane with two MWCO could effectively eliminate bacterial endotoxins, but the content of effectual components was significantly decreased after ultrafiltration by membrane with MWCO 10 kDa,and the reservation of effectual components was above 95% by ultrafiltration membrane with MWCO 50 kDa. Ultrafiltration membrane,with cut off molecular weight 50 kDa,which can effectively eliminate bacterial endotoxins and reserve effectual components, are applied to used in the production of Xuesaitong injection.
