摘要
应用微管吸吮技术测量了烧伤血清剌激后24h内中性粒细胞(PMN)与内皮细胞(EC)粘附力的变化,并应用流式细胞术(FCM)检测了烧伤血清作用下介导PMN-EC粘附的主要3个粘附分子:CD11a/CD18、CD11b/CD18和细胞间粘附因子-1(ICAM-1),以探讨PMN-EC粘附力的分子基础。发现PMN在烧伤血清剌激后与正常体外培养的人脐静脉内皮细胞(HUVEC)间粘附力迅速上升,于1h达到饱和点,并在12h内保持此水平。烧伤血清剌激HUVEC后与未受剌激的PMN间粘附力逐渐升高,至24h达峰值,在以后12h内维持此水平。烧伤血清剌激后,PMN其膜表面的CD11a/CD18、CD11b/CD18迅速表达增强,1h达峰值,在24h内维持此高表达水平;而HUVEC受烧伤血清剌激后ICAM-1表达逐渐增多,至12h达最大值,在以后12h无明显变化。提示:烧伤血清可剌激PMN和EC分别表达CD11a/CD18、CD11b/CD18和ICAM-1,从而加强PMN-EC粘附。
This study was to investigate the change of adhesive force between polymorphonuclear neutrophils (PMN) and endothelial cell(EC) in response to burnt patients' serum and its mechanisms in vitro. Adhesive force between a PMN and a HUVEC was calculated by micropipette technique, expressions of CD11a/CD18, CD11b/CD18 and ICAM1 were determined by flowcytometry. The findings were as follows: Adhesive force between a PMN and a HUVEC increased sharply after PMN incubated with burnt serum, which reached the maximum level at 1h after incubation and remained this level in the following 23. However, after HUVEC was stimulated by burnt serum, PMNHUVEC adhesive force increased gradually, reached peak at 12h and remained this level in following 12 h. In response to burnt serum, expressions of CD11a/CD18 and CD11b/CD18 on PMN increased, reached the peak at 1h, and maintained this level within 24 h; expression of ICAM1 on HUVEC increased gradually, reached the peak at 12h. The results suggested that burnt serum could enhance adhesive force between PMN and EC, stimulate expressions of CD11a/CD18,CD11b/CD18 and ICAM1.Therefore,burnt serum played a great role in PMNEC adhesion after burn injury. CD11a/CD18, CD11b/CD18 and ICAM1 were the molecular base of PMNEC adhesion.
