普伐他汀对心脏成纤维细胞转化生长因子βⅠ型受体mRNA表达的影响

Effects of pravastatin on expression of transforming growth factor-beta receptor type Ⅰ mRNA in cardiac fibroblasts

摘要 目的观察普伐他汀对醛固酮诱导新生大鼠心脏成纤维细胞转化生长因子βⅠ型受体(TGF-βRI) mRNA表达的影响及其机制。方法采用胰酶消化法和差速贴壁分离法获取新生大鼠心脏成纤维细胞,应用RT-PCR的方法测定TGF-βR I mRNA的表达。结果醛固酮(0.1μmol·L^(-1))可诱导心脏成纤维细胞中TGF-βR I mRNA的表达,预先24 h给予普伐他汀(0.01、0.1、1 mmol·L^(-1))能剂量依赖性抑制醛固酮的上述作用,同时,普伐他汀的这种抑制作用可被甲羟戊酸所逆转。结论普伐他汀可抑制醛固酮诱导的心脏成纤维细胞TGF-βR I mRNA表达,其机制可能与甲羟戊酸代谢途径有关。
