乙型脑炎病毒E基因自杀性DNA疫苗的构建及其免疫应答 被引量:3

摘要 为探讨自杀性DNA疫苗在乙型脑炎疫苗开发中的可能性,将乙型脑炎病毒(JEV)E基因片段插入到自杀性DNA疫苗载体pSCA1(pS)中,构建了表达E基因的自杀性DNA疫苗质粒pSE。间接免疫荧光试验结果证实,该质粒转染BHK-21细胞后实现了E基因在细胞中的表达。将该疫苗和常规DNA疫苗(pCDE)分别免疫试验小鼠,进行体液免疫和细胞免疫检测。结果表明,在诱导体液免疫方面,pCDE要优于pSE,但pSE却可以诱导更强的细胞免疫反应,这为进一步评价该疫苗的免疫保护效果奠定了基础。 To evaluate the possibility of the suicidal DNA vaccine against Japanese encephalitis virus (JEV) ,a suicidal DNA vaccine plasmid(pSE) expressing E gene of JEV was constructed by inserting the E gene fragment into the expression vector pSCA1. The plasmid was then transfected into BHK-21 cells and the E protein expression was demonstrated by indirect immunofluorescenee assay(IFA). A DNA vaccine expressing the same E gene(pCDE) was used as a control to immunize mice. Then the humoral and cellular immunity responses were detected and compared with each other. The results revealed that pCDE was significantly superior to pSE in humoral immune response,but the cellular immune response level induced by pSE was significant higher than that of pCDE. So the study laid a foundation for evaluate the protective efficacy of pSE.
