摘要
U937细胞、经PMA及dbcAMP预处理的U937细胞,用rhC5a、PMA、A23187分别诱导,以ELISA法测定细胞培养上清中IL-8,证实:rhC5a不能诱导U937细胞产生IL-8,但可诱导经PMA、dbcAMP预处理的U937细胞产生IL-8;用PMA激活PKC和(或)以A23187提高细胞内Ca2+浓度可使IL-8分泌水平增高。研究结果有助于加深对补体系统与细胞因子之间关系的认识,以及探索减轻C5a致病作用新途径。
he interleukin8 in the supernatants of U937 cells,PMA or dbc AMP pretreated U937 cells which were induced with rhC5a,PMA and A23187 were detected by ELISA.Stimulated with rhC5a,U937 didn′t secrete detectable IL8,but PMA or dbc AMP pretreated U937 cells secreted detectable IL8.It was discovered that activation of PKC with PMA and(or) rise in cytosolic Ca2+ with A23187 increased secretion of IL8.The result contributed to finding a new way to reduce the pathogenesis of C5a and getting a deeper understanding of the relationship between complement system and cytokines.
