摘要 U937细胞、经PMA及dbcAMP预处理的U937细胞,用rhC5a、PMA、A23187分别诱导,以ELISA法测定细胞培养上清中IL-8,证实:rhC5a不能诱导U937细胞产生IL-8,但可诱导经PMA、dbcAMP预处理的U937细胞产生IL-8;用PMA激活PKC和(或)以A23187提高细胞内Ca2+浓度可使IL-8分泌水平增高。研究结果有助于加深对补体系统与细胞因子之间关系的认识,以及探索减轻C5a致病作用新途径。 he interleukin8 in the supernatants of U937 cells,PMA or dbc AMP pretreated U937 cells which were induced with rhC5a,PMA and A23187 were detected by ELISA.Stimulated with rhC5a,U937 didn′t secrete detectable IL8,but PMA or dbc AMP pretreated U937 cells secreted detectable IL8.It was discovered that activation of PKC with PMA and(or) rise in cytosolic Ca2+ with A23187 increased secretion of IL8.The result contributed to finding a new way to reduce the pathogenesis of C5a and getting a deeper understanding of the relationship between complement system and cytokines.
