海水培养光合细菌存在的问题及其优化研究 被引量:4

摘要 [目的]为光合细菌海水培养技术的大规模应用提供理论依据。[方法]采用对比试验研究光合细菌在不同培养基中的生长状况,探讨光合细菌在海水培养过程中出现沉降现象的原因,在此基础上对培养基组分进行优化并进行优化培养试验。[结果]在海水培养光合细菌的过程中,酵母膏、蛋白胨培养基中沉降现象较轻,其余培养基中培养2 d后均出现较严重的沉淀现象,主要是因为培养过程中培养基的pH值逐渐升高,其中的磷酸盐与海水中的Ca2+和Mg2+结合成沉淀,从而使细菌附着在沉淀表面生长所致。优化后的培养基组分为:乙酸钠3.0 g/L,氯化铵1.0 g/L,磷酸二氢钾0.1 g/L,酵母浸膏0.5 g/L。[结论]用优化后的培养基培养光合细菌效果理想,培养7d后光合细菌的个数可达3.16×109个/ml。 [Objective] The aim was to provide theoretical basis for the application of culture technology for photosynthetic bacteria in seawater in large scale. [Method] The contrast test was taken to study the growth status of photosynthetic bacteria in different mediums and to explore the reason of photosynthetic bacteria appearing subsidence phenomenon in seawater in culture process. On the basis of it, the medium components were optimized and the optinfized culture was carried out. [Result] In the process of photosynthetic bacteria culture in seawater, the subsidence phenomenon in yeast extract and peptone medium was slight and that in other mediums was serious after cultured for 2 d. The main reason was that, with the gradually rise of pH value of medium in culture process, the phosphate combined into precipitation with Ca^(2+) and Mg^(2+) in seawater, so the bacteria attached to the precipitation to grown.. The optimized medium was that sodium acetate of 3.0 g/L, chloride of 1.0 g/L, potassium dihydrogen phosphate 0.1 g/L, yeast extract 0.5 g/L. [Conclusion] The culture effect of ohotosynthetic bacteria with optimized medium was ideal, and the number of photosynthetic bacteria could reach 3.16×10^9 ind/ml after cultured for 7 d.
