日本血吸虫细胞体外培养实验研究

Experimental study of Schistosoma japonicum cell cultivation in vitro

摘要 收集血吸虫毛蚴、尾蚴和成虫,无菌条件下进行组织处理,接种于含有胎牛血清、双抗、促细胞生长物的RPMI-1640培养基中进行细胞原代及传代培养。结果日本血吸虫细胞体外培养及传代获得成功,组织原代培养可见在组织碎块周围有发亮、饱满,呈单个分布或索状排列的细胞;随时间的延长,显示细胞增殖。原代培养细胞长满单层后,进行传代接种,传代后细胞形成密集、大小相近、分布均匀。 Collect schistosome myracidium, cercariae and imago to perform organic process under aseptic condition, and vaccinate into RPMI 1640 culture medium containing fetal bovine serum, penicillin and streptomycin that promote cell growth substance for primary cell cultivation and sub-cultivation. Results indicated the success of in vitro Schistosoma japonicum cell cultivation and sub-cultivation. Primary organic cultivation showed full of individual or funicular distributed shiny cells surrounding organic fragment periphery and proliferating along with time extension. After the cultivated primary cells covered with monolayer, passage vaccination occurs. Once completing passage cultivation, the cells were dense formed, similarly sized and evenly distributed.
