γ-射线照射梨试管苗诱导产生多倍体变异 被引量:9

Polyploid Induction in Pear in vitro Treatment with Gamma-rays

摘要 用γ-射线照射梨试管苗,照射后对其顶芽、侧芽和叶片3种外植体分别接种,经过继代和筛选,获得了形态变异明显的变异无性系,其变异表现为茎变粗,节间变短,叶长/叶宽比变小,叶柄变短。经染色体计数鉴定,这些变异中既有三倍体也有四倍体。多倍体的气孔长度及比叶重较对照二倍体显著增大。 In vitro shoots of pear cultivar ' Fertility' (Pyrus communis L. ) were irradiated with gammarays. Three different explants of shoot tip, lateral bud and young leaf from irradiated shoots were cultured separately. Shoots from 3 different explants were subcultured and selected. Morphological mutants with thicker stem, shorter internode, smaller leaf index and shorter petiole were obtained. Chromosome counting confirmed that the mutants were triploids and tetraploids. The lengths of stomatal guard cells and specific leaf mass of polyploids were significantly increased than that of diploids.
