摘要
目的探讨胰高血糖素样肽-2(GLP-2)对急性胰腺炎(AP)小鼠肠道淋巴细胞归巢受体整合素α4β7和归巢配体肠道黏膜地址素细胞黏附分子-1(MAdCAM-1)表达的影响。方法将96只小鼠随机分为AP组、GLP-2组、对照组,每组32只。运用雨蛙素联合脂多糖腹腔注射制备小鼠AP模型。GLP-2组于制模后15min开始腹腔注射250μg/kg GLP-2,每12h1次,连用3d;对照组给予等量生理盐水。分别于制模后6、12、24、48h处死小鼠后取标本,流式细胞仪检测外周血中整合素α4β7的阳性淋巴细胞数,免疫组化法测定末端回肠和Peyer结中MAdCAM-1的表达。结果AP组小鼠制模后各时间点外周血中整合素α4β7阳性淋巴细胞数及末端回肠和Peyer结中MAdCAM-1表达均较对照组明显降低(P均〈0.05);应用GLP-2后整合素α4β7阳性淋巴细胞数及MAdCAM-1表达均较AP组明显升高(P均〈0.05),但仍低于对照组(P均〉0.05)。结论GLP-2能改善AP小鼠整合素α4β7和MAdCAM-1的表达,促进肠道淋巴细胞归巢,从而改善AP时的肠道免疫功能。
Objective To investigate the influence of glucagon-like peptide-2 (GLP-2) on intestinal lymphocyte homing receptor-integrin α4β7 and homing ligand-intestinal mucosal addressin cell adhesion molecule-1 (MAdGAM-1) in mice with acute pancreatitis (AP). Methods A total of 96 mice were divided into three groups randomly (n= 32 in each group): AP group, GLP-2 group and control group. Murine AP model was reproduced by intraperitoneal injection of caerulein and lipopolysaccharides (LPS). GLP-2 (250 μg/kg) was injected intraperitoneally at 15 minutes after the establishment of model, then it was injected twice a day for 3 days in GLP-2 group, while the mice in control group received normal saline instead. Mice were sacrificed at 6, 12, 24 and 48 hours after reproduction of AP, and tissue specimens were harvested. Integrin α4β7 positive peripheral blood lymphocytes were determined by flow cytometry. The expression of MAdCAM-1 in the terminal ileum mucosa and Peyer patch was measured by immunohistochemistry. Same observations were also done in the control and GLP-2 groups. Results Compared with control group, integrin α4β7 positive lymphocyte in peripheral blood and the expression of MAdCAM-1 in the terminal ileum mucosa and Peyer patch were significantly reduced at 6, 12, 24 and 48 hours in AP mice (all P〈0. 05). Integrin α4β7 positive lymphocyte and the expression of MAdCAM-1 were markedly higher in GLP-2 group than those in AP group (all P〈0.05), but were lower in GLP-2 group than those in control group (all P〈 0. 05). Conclusion Administration of GLP-2 may restore expression of integrin α4β7 and MAdCAM-1, promote lymphocyte homing to intestine, thus improve the immunological function of intestine.
