成肌细胞移植治疗DMD模型-mdx鼠骨骼肌Dystrophin表达的实验研究 被引量:3

摘要 目的探讨成肌细胞移植(MTT)治疗Duchenne型肌营养不良症模型-mdx鼠的可能性。方法运用体外分离培养6d处在分裂增殖期的成肌细胞,经过纯化并以FG荧光标记后进行肌肉注射于Duchenne型模型鼠-mdx鼠。移植后1月、2月、3月分别取实验鼠的四肢骨骼肌,运用免疫荧光法检测抗肌萎缩蛋白的表达情况。结果成肌细胞移植后1,2,3月后mdx鼠的抗肌萎缩蛋白阳性肌纤维增加。结论成肌细胞肌肉注射移植能够在一定程度上修复mdx鼠的萎缩肌肉组织。 Objective To explore the curative ability of myoblast transfer therapy (MTT)for Duchenne muscular dystrophy model-mdx mice. Methods The myoblasts which were labelled by FG fluorochrome was transplanted into mdx mice by intramuscular injection. At the 1st, 2ed, 3rd month after MTT by intramuscular injection, to observe dystrophin expression in skeletal muscle of mdx mouse. Results 1-3months after implantation, Dystrophin protein expressing increased obviously, the mosaic myofibers simultaneously labeled with FG increased obviously. Conclusion There is some restoration function for myopathy in mdx mouse by MTT.
