摘要
应用胶体金免疫层析技术建立了一种快速检测莱克多巴胺的方法。采用柠檬酸钠还原法制备胶体金颗粒,标记莱克多巴胺单克隆抗体并喷于玻璃纤维上,莱克多巴胺偶联OVA抗原和羊抗鼠IgG分别结合于硝酸纤维膜上,依次将样品垫、胶体金垫、硝酸纤维素膜和吸水纸组装切割成莱克多巴胺胶体金免疫层析快速检测试纸条。试验结果表明,该快速检测试纸条的灵敏度为5 ng/mL,检测时间为5 min,批内和批间重复性为100%,假阳性率和假阴性率均为0。
To establish a rapid assay for detection of ractopamine, gold immunochromatography assay (GICA) was used. Colloidal gold marked with monoclonal antibody against ractopamine was sprayed onto the glass fiber. Ractopamine-OVA and goat antimouse immunoglobulins were jet-positioned onto anitrocellulose membrane. Sample pad, gold jet-pad, nitrocellulose membrane and absorbent paper were, assembled and cut into detecting card, respectively. The results showed that the visual detection limit for the rapid assay of ractopamine is 5 ng/mL, detection time is within 5 minutes and repeatability of the strip is 100%. The assay gives no false positive and false negative results.
