目的观察NF-κB激活和抑制对血管内皮细胞凋亡的影响。方法获取和培养原代脐静脉血管内皮细胞(HUVEC),分别以NF-κB-decoy和错配-decoy预处理细胞,然后以内毒素LPS刺激诱发内皮细胞凋亡,设立单一LPS处理组和空白组做对照,比较各组之间NF-κB活性和凋亡率差异。结果LPS显著增加血管内皮细胞凋亡,NF-κB decoy组NF-κB活性被显著抑制(P<0.05),而错配decoy组则不受影响;LPS组,错配decoy组和NF-κB decoy组凋亡率依次递减,三组之间P<0.05。结论NF-κB激活可促进血管内皮细胞凋亡,抑制其活性可能减轻由于内皮细胞凋亡带来的毛细血管渗漏。
Objective To observe the influence of nuclear factor-κB (NF-κB) activation and suppression on human umbilical vein endothelial cells apoptosis. Methods Human umbilical vein endothelial cells were isolated from fresh human umbilical cord veins. After NF-κB decoy or scramble decoy was transfected, endothelial cells were stimulated with LPS at a final concentration of 25 μg/ml. Then the discrepancy of nucleolus NF-κB activity and cell apoptosis rate between groups were compared. Results LPS stimulation increased endothelial cell nucleolus NF-κB activity and cell apoptosis. The increment of NF-κB activity was significantly ameliorated in NF-κB decoy group, but not in scrambled decoy group. Endothelial cells apoptosis increment in the presence of LPS stimulation was abrogated by NF-κB decoy. Conclusion NF-κB activation could promote endothelial cells apoptosis and the effect could be suppressed by NF-κB decoy strategy. This indicated that NF- κB activity suppression strategies might be applied to capillary leakage syndrome treatment.
