摘要
采用PCR—RFLP技术,分析脂肪酸合成酶(FAS)基因启动子区在朗德鹅中的AfaI酶切片段多态性分布。FAS基因启动子区具有3个AfaI酶切位点(位点A、B和E),3个位点在朗德鹅中均具有多态性。通过χ^2检验显示3个酶切位点都不符合哈代-温伯格法则。最小二乘分析结果显示:E位点中FAS^EE基因型个体在活体质量、填饲期增质量、屠体质量、半净膛质量、全净膛质量、胸肌质量、腿肌质量方面比FAS^ee基因型个体高出12.78%~19.67%(P〈0.05);位点B具有类似的影响趋势;而位点A对屠宰性能没有显著影响(除了胸肌质量)(P〉0.05)。在3个酶切位点中,位点A和B的杂合基冈型FAS^Aa和FAS^Bb个体产肝性能比纯合基因型好,而E位点的FAS^EE基因型个体肝质量比FAS^EE基因型个体高33.09%(P〈0.05)。3个酶切值点对体脂沉积性状影响均不显著。
The aim of this study was to investigate polymorphism of fatty acid synthase (FAS) gene promoter region in Landes goose. Polymorphisms were detected by restriction fragment length polymorphisms(RFLP) and enzyme Afa I was used. Three Afa I digestion sites (locus A, B and E) were detected in Landes goose. The χ^2-test was performed to examine the Hardy- Weinberg equilibrium (HWE). Three sites significantly deviated from HWE. The least square a- nalysis showed that living weight, weight gain at feeding increase stage, carcass weight, halfevis- cerate weight, eviscerate weight, breast muscle weight and leg muscle weight of individuals with genotype FASEE were higher than that of individuals with genotype FAsee12.78%-19.67o%(P〈0.05) at loci E and B. Locus A had no significant effects on carcass traits except for breast muscle weight(P〉0.05). Fatty liver performance of individuals with FAS^Aa and FAS^Bb were better than that of individuals with homozygote at loci A and B, but the liver weight of individuals with genotype FAS^EE was higher than that of individuals with heterogenesis 33.09%(P〈0.05)at locus E. Three loci had no significant effects on fat deposit traits.
