摘要
目的:首次探讨激光爆破脂质体微泡造影剂的可行性,并研究其对视网膜母细胞瘤细胞增殖的影响。方法:利用激光在液体中的光声效应,采用Nd-YAG激光,将激光聚焦于脂质体微泡,按照不同的能量参数分组照射,通过DFY-Ⅱ型超声定量分析诊断仪检测微泡爆破率;台盼蓝染色及MTT法研究其对视网膜母细胞瘤细胞增殖影响。结果:激光可成功爆破微泡,不同单脉冲能量(5mJ、10mJ、15mJ)、照射总能量(125mJ、250mJ、500mJ、750mJ)之间相比有统计学意义(P<0.05);不同激光微泡参数对视网膜母细胞瘤细胞增殖影响亦具统计学意义(P<0.05);结论:Nd:YAG激光能够成功爆破脂质体微泡,在合适的参数下,对视网膜母细胞瘤细胞增殖无明显影响。
Objective:To investigated the feasibility of using laser to dynamite liposome microbubble contrast medium and the influence to retinoblastoma cell proliferation at the first time. Methods: After the preliminary test, we chosen the Nd - YAG laser. The laser light focused on liposome microbubble contrast medium, cells suspl and liposome microbubbje contrast medium/cells suspl, the energy of the light were different in each group. The feasibility of using laser to dynamite liposome microbubble were evaluated by the dynamite rate of the microbubble detected by DFY - 11 ultrasonic diagnosis analysator, and the influence to retinoblastoma cell proliferation was dectected by Trypan blue staining method and MTT. Results: It was found that different single pulse energy (5mJ, 10mJ, 15m J), different total energy ( 125mJ ,250mJ,500mJ, 750mJ) had different dynamite rate, the difference was markedly in statistic (P 〈 0. 05) , and so the influence to retinoblastoma cell proliferation. Conclusion: It was regarded that the Nd- YAG laser could successfully dynamited the liposeme microbubble contrast medium; if the total energy lower than 500mJ, the Influence to retinoblastoma cell proliferation has no significance.
