实时定量大肠癌组织中MK基因表达及其临床意义

摘要 目的利用实时定量PCR(Real-time Quantitative Polymerase Chain Reaction,RT—PCR)相对定量大肠癌组织中Midkine(MK)基因的表达,探讨MK基因在大肠癌不同病理状态中的意义。方法提取34例临床大肠癌组织及其癌旁正常组织中的总RNA,经寡聚脱氧胸腺嘧啶逆转录,RT—PCR扩增,以内标甘油醛-3-磷酸脱氢酶(GAPDH)基因的mRNA相对定量MK基因的mRNA表达,比较癌组织与其癌旁正常组织中的MK表达差异。结果34例样本中癌组织较之癌旁正常组织,MK基因表达降低(P〈0.01);其中在Duckg分期中C期、女性组、淋巴结转移、管状腺癌和浸润浆膜层组中癌组织较之癌旁正常组织表达降低(P〈0.05),其余组大肠癌组织较之癌旁正常组织无统计学意义(P〉0.05)。结论在大肠癌病程晚期癌组织中MK基因的表达水平降低。 Objective With Real-time Quantitative Polymerase Chain Reaction (RT- PCR) Midkine (MK) gene expression was relatively quantified in colorectal cancer sanaples. We discussed a relationship of MK expression and colorectal cancer. Methods The total RNA was extracted from 34 clinical samples of colorectal cancerous tissue and their adjacent normal tissue. Tne RNA was reverse transcripted into cDNA with oligodT. And the cDNA was amplified by RT- PCR. MK relative expression was quantified to an inherence gene Glyceraldehyde - 3 - phosphate (GAPDH). At last the different was analyzed between colorectal cancerous tissue and their adjacent normal tissue. Result In the 34 samples MK mRNA of the colorectal cancerous tissue is less than that of the adjacent normal tissue ( P 〈 0.01 ). And the same situation is MK mRNA in thel4 C - phase samples, the female, the transferred lymphaden, the tubular adenocareinoma and the invasive subserous groups ( P 〈 0.05). But there is no statistical signification ( P 〉 0.05) in the other groups. Conclusion MK expresses lowly in the eolorectal cancer of late stage.
