摘要
目的研究来曲唑诱导建立多囊卵巢综合征(PCOS)动物模型中卵巢组织瘦素受体(Ob-R)及其mRNA的表达,探讨卵巢Ob-R变化与PCOS发病机制的关系。方法6周龄清洁级雌性SD大鼠45只,分为正常组、溶剂组和来曲唑组,各15只。正常组每日自由摄取食物和水;溶剂组每日给予1%羟甲基纤维素水溶液2 mL/kg灌胃;来曲唑组以来曲唑0.5 mg/kg灌胃。观察大鼠体质量、动情周期、卵巢质量及其形态学改变,测定血清黄体生成素(LH)、睾酮(T)、胰岛素(INS)、瘦素(LEP)及血糖(FG)水平,采用免疫组织化学和Real Time RT-PCR方法检测大鼠卵巢中Ob-R及其mRNA的相对表达,并对3组的各指标进行比较。结果来曲唑组大鼠体质量和卵巢质量指数升高,动情周期消失,卵巢体积增大,白膜增厚,H-E染色可见较多囊状扩张的卵泡且黄体数量明显下降,血清T、LH、INS、LEP水平明显升高(P<0.05);卵巢Ob-R及其mRNA表达明显增强(P<0.05)。结论来曲唑诱导建立的PCOS动物模型临床表现与人类似;卵巢中Ob-R表达上调,可能是导致PCOS患者不排卵、生育力下降的原因之一。
Objective To investigate leptin receptor in the ovaries of rat model with polycystic ovarian syndrome (PCOS) induced by Letrozole, in relation to pathogenesis of PCOS. Methods Fortyfive female SD rats of 6 weeks old were divided into normal group, solvent group and letrozole group. The letrozole group was gavaged with Letrozole (0. 5 mg/kg). Observation and comparison for body weight, estrous cycle, ovarian weight and ovarian morphological changes were conducted. Serum T,LH, FG,INS, and LEP level were measured, The relative expression of leptin receptor in the ovaries was measured by immunohistochemistry and Real Time RT-PCR. Results The body weight and ovarian weight in model group were higher than those in normal control group and solvent control group (P〈0.05). Estrous cycle disappeared, ovarian volume increased and tunica albuginea went thickening in model group. With H-E staining, visible cysts significantly increased and the ratio of corpus luteum decreased. Serum testosterone, luteinizing hormone, insulin, and leptin levels in model group were higher than those of the other two groups. The expression of leptin receptor in the ovaries of model group was higher than that of the other two groups (P〈0.05). Conclusion The Letrozole-induced PCOS in rat models is similar to that in patients. The Increasing expression of leptin receptor in the ovary may lead to no ovulation and declining fertility of the patients with PCOS.
