摘要 目的:建立一种新的荧光法测定尿样中痕量锰。方法:在pH8.2的H3BO3-NaOH缓冲介质中,Mn2+与1-(2-吡啶偶氮)-2萘酚(PAN)形成配合物;在乳化剂OP胶束溶液中,λex/λem=539/553 nm,曙红Y(EY)与PAN-Mn2+配合物之间发生能量转移,使EY的荧光猝灭,在一定范围内荧光猝灭值与Mn2+浓度成正比,由此建立测定痕量锰的荧光分析新方法。结果:在选定实验条件下,Mn2+含量在8.2~600 ng/ml范围内与荧光猝灭程度呈现良好的线性关系(r=0.9999)。方法的检出限为2.45 ng/ml;相对标准差为0.33%~1.08%(n=11);样品加标回收率为98.9%~105.4%。结论:方法加入试剂简单,稳定性好,灵敏度高,操作简便,用于尿样中痕量锰的测定,结果满意。 Objective:To set up a new fluorescence method for the determination of trace Mn2+ in urine. Methods:In the H3 BO3 - NaOH buffer solution at pHS. 2, Mn2+ and PAN interacted to form a complex. An effective energy transfer could occur between Eosin Y (EY) and PAN -Mn2+ complexes at the condition of λex/λem =539/553 nm and in the presence of emulsifying agent OP, which led to the great decrease of fluorescence intensity of EY. Based on this phenomenon, a novel method of fluoremetric energy transfer for determination of trace Mn2+ was established. Results:There was a good linearity between fluorescence quenching intensity and mass concentration of Mn2+ in the range of 8. 2 -600 ng/ml( r = 0. 9999 ). The detection limit for Mn2+ was 2.45 ng/ml. The relative standard deviation (RSD) was 0. 33% - 1. 08% ( n =11 ) and the sample average recoveries were 98.9% - 105.4%. Conclusion :The method exhibits good recurrence, high sensitivity and simplicity of operation. It can be satisfactorily applied to the determination of trace amount of Mn2 + in urine samples.
