摘要
目的:评价当归-川芎不同配比药对挥发性成分与其抑制小鼠离体子宫平滑肌收缩效应的相关性。方法:采用小鼠离体子宫收缩模型评价当归-川芎不同配比药对挥发性成分的生物效应,采用气相色谱-质谱(GC-MS)联用技术对挥发性成分进行分析鉴定,探讨两者的相关性。结果:离体子宫平滑肌收缩试验表明不同配比当归-川芎药对挥发油对小鼠离体子宫收缩效应强弱依次为:当归-川芎(2∶1)>当归-川芎(3∶1)>当归-川芎(1∶1)>当归-川芎(3∶2);GC-MS分析结果表明对离体子宫收缩效应最佳的当归-川芎(2∶1)药对挥发油中主要成分的比例关系为:(Z)-ligustilide:(Z)-3-butylidene phthalide:4,5-dihydro-3-butyl-phathalide:(E)-ligustilide:Unknown compound:8,9-dehydro-Cycloisolongifolene:Neocnidilide:(E)-3-butylidene phthalide(80.75∶4.02∶2.07∶1.66∶0.81∶0.61∶0.53∶0.52)。结论:不同配比当归、川芎药对挥发性成分抑制离体子宫平滑肌收缩效应强度不同,可能是由于挥发油中各组成成分比例不同引起;挥发油中以内酯类成分为主,提示该类成分可能为抑制子宫收缩的主要活性成分。
contraction in vitro Objective: To analysis the and volatile components of correlation between bioactivity of inhibiting oxytoein-induced uterine Danggui-Chuanxiong different ratio. Methods: The essential oil was extracted from Danggui-Chuanxiong by steam distillation and analyzed by GC-MS with the method of programmed temperature. Oxytocin induced female mice isolated uterine contraction model was adopted in the experiment to evaluate biological activity of Myrrh oil. Results: The experiments showed that the activities of volatile oils obtained from different ratio Danggui-Chuanxiong were different : Danggui-Chuanxiong (2 : 1 ) 〉 Danggui-Chuanxiong ( 3 : 1 ) 〉 Danggui- Chuanxiong (1 : 1 ) 〉 Danggui-Chuanxiong (3 : 2) ; The results of GC-MS showed that the constituents of volatile oil obtained from Danggui-Chuanxiong (2 : 1 ) were composed with the ratios of ( Z )-ligustilide : ( Z )-3-butylidene phthalide: 4,5-dihydro-3-butyl- phathalide : ( E)-ligustilide : Unknown compound : 8,9-dehydro-Cycloisolongifolene : Neocnidilide : ( E)-3-butylidene phthalide (80.75 : 4.02 : 2.07 : 1.66 : 0.81 : 0.61 : 0.53 : 0.52). Conclusion : The different activities of volatile oils derived from different ratio of Danggui-Chuanxiong were resulted from the different composition of components in volatile oils. And the results suggested that the lactone-type components are possible bioactive compounds.
