摘要
运用流式细胞仪及细胞原位杂交方法测定低氧对培养的新生牛肺动脉平滑肌细胞(PASMC)生长的影响,并探讨血小板源生长因子(PDGF)在其中的作用。结果表明:培养的PPAMC在低氧12h时,由G/G期进入S期的细胞数明显增多,为对照组的157.7%(P<0.05):PDGF可进一步促进低氧状态下PASMC的增殖,S期细胞为对照组的242.3%(P<0.01)。原位杂交结果显示:低氧(12h)条件下,PDGF-A.B链及a、b受体基冈表达量显著增高(P<0.001)。提示:低氛条件下的PDGF及其受体基因表达增强与低氧的促增殖作用有协同效应,在低氧性肺血管重经过程中,可能是参与PASMC肥大、增生机制之一。
By means of flow cytometry and in situ hybridization ,we investigated the role of hypoxia in cultured neonatal bovine pu,lmonary artery smooth muscle cells (PASMC ) ,and the effect of PDGF on that role. Cell-cycle analysis revealed hypoxia ( 12h ) and PDGF added meantime had effect on the G/G S transition , both could increase the number of cells entering S phase from G/G The percents of S phase cells reached to 41%and 63%respectivly , and werel 57. 7% and 2.2. 3% compared with normoxiit (P<0. 05). Using in situ hybridization , we detected that hypoxia (12h ) upregulated PDGF-A,B chain and receptor gene expres- sion (all P<0. 001). Results therefore suggested that increase of PDGF-A ,B chain and receptor gene elxpression under hypoxia condition and effect of PDGF cooperated with hypox- ia on PASMCs proliferation may contribute to the pulmonary vascular remodeling.
