银杏总黄酮和银杏内酯配伍对心肌缺血猫心外膜心电及心肌SOD的影响 被引量:4

Effects of total flavone of Ginkgo combined with Ginkgolides on ECG of epicardium and activity of SOD on myocardial ischemia in cats

摘要 【目的】初步探索不同比例银杏总黄酮和银杏内酯伍用对猫心肌缺血的保护作用。【方法】将35只家猫随机分为7组,银杏总黄酮组、银杏内酯组、总黄酮与内酯比例分别为3∶1、1∶1、1∶3组,生理盐水组和美托洛尔组,给药剂量为0.5mg生药/kg。将家猫开胸,结扎左冠脉前降支造成局部心肌暂时缺血模型,测定结扎前和造模后第1、3、5、10、15min各时间点缺血区心外膜心电图ST段高度;恢复供血后给药,再次结扎,重复上述时间点ST段测定;t检验自身对照比较给药前后心肌缺血ST段变化。术后剪取缺血区心肌测定其SOD活性;对缺血心肌抗氧化能力进行方差分析。【结果】银杏总黄酮与银杏内酯比例为3∶1时,抑制心外膜心电图ST段抬高的效果最明显,且心肌组织SOD活性最高。【结论】在本实验条件下,银杏总黄酮与银杏内酯比例为3∶1时,能较好改善猫心肌缺血的症状。 [ Objective] To investigate the protection of different ratios between Total Flavone of Ginkgo (TFG) and Ginkgolides used on myocardial ischemia in cats. [ Methods] 35 Cats were divided into 7 groups randomly, every cat was given medicine at the dose of 0.5mg/kg. They were TFG group, Ginkgolides group, TFG: Ginkgolides at 3 : 1 group, TFG: Ginkgolides at 1 : lgroup, TFG: Ginkgolides at 1:3 group, model group and metaprolol group. Operation was had to ligate the left anterior descending (LAD), built the model of acute myocardial ischemia in cat. The ST segment of ECG of epicardium was measured before ligate the LAD and at the time of the 1st, 3rd, 5th, 10th, 15th minute after ligated the LAD in model cat; then loosened it to measure the height of ST segment of recovery, gave medicine and measured the height of ST segment after ligated the LAD all the moments above; test t of paired design use self-contrast. At last, measured the activity of Superoxide dismutase (SOD) of ischemic myocardium after the operation; the antioxidation of ischemie myocardium of statistic design use analysis of variance. [Results] TFG: Ginkgolides at 3:1 group show the best effect to inhabit the ST segaent of ECG of epicardium, and the activity of SOD of ischemie myocmdium is highest in this group. [ Conclusions] TFG: Ginkgolides at 3:1 group can protect the ischemic myocardium most effectively in this condition.
