摘要
为确定绵羊防御素(Sheep beta defensin-1,sBD1)mRNA在组织器官中的分布和不同发育阶段的表达是否存在差异。利用RT-PCR(Reverse Transcription PCR)方法检测绵羊防御素sBD1 mRNA在组织器官的分布情况,通过荧光定量RT-PCR(Fluorescence quantitative RT-PCR)检测不同发育阶段绵羊防御素的表达差异。结果表明:在胎羊的舌、皱胃、十二指肠、空肠、盲肠检测到绵羊防御素sBD1 mRNA,在羔羊和成年羊消化道的舌、食道、瘤胃、皱胃、十二指肠、空肠、回肠、盲肠、直肠和呼吸道的气管、肺均检测到绵羊防御素sBD1 mRNA,在各发育阶段的心、肝、肾以及羔羊、成年羊的淋巴结和体外培养的羔羊肺泡巨噬细胞均未检测到sBD1 mRNA;实时定量PCR比较表明,sBD1 mRNA在不同发育阶段皱胃、十二指肠和空肠表达量的高低为sBD1 mRNA表达量(羔羊)>sBD1 mRNA表达量(成年羊)>sBD1 mRNA表达量(胎羊)。绵羊防御素sBD1 mRNA主要分布在消化道和呼吸道,且在整个发育过程中是持续表达的,其在消化道表达量高低为sBD1 mRNA表达量(羔羊)>sBD1 mRNA表达量(成年羊)>sBD1 mRNA表达量(胎羊)。
Detecting distribution and expression level of sheep beta-defensinl in differently developmental stages. Sheep beta-defensin1 mRNA was detected by traditional RT-PCR and fluorescence quantitative RT-PCR. sheep beta-defensinl was mainly expressed in digestive and respiratory tracts, included tongue, esophagus, rumen, abomasum, duodenum, jejunum, ileum, colon, appendix,trachea and lung, but not in heart,liver,kidney,lymph node and alveolar macrophage. The expression level of beta-defensinl in abomasum,duodenum and jejunum was sBD1 mRNA expression level (neonatal)〉sBD1 mRNA expression level(adult))sBD1 mRNA expression level(fetal sheep). Sheep beta-defensinl was consistently expressed in development process and its expression level in gastrointestinal tract was sBD1 mRNA expression level (neonatal)〉sBD1 mRNA expression level(adult)〉sBD1 mRNA expression level(fetal sheep).
