摘要 目的研究体外建立模拟CO2、He气腹环境对胃癌MKN-45细胞增殖的影响,初步探讨CO2气腹腹腔镜胃癌手术的安全性。方法体外建立模拟CO2、He气腹环境,全自动气腹机维持压力为12mmHg,作用3h并没对照组。处理后0、0.5、1、1.5、2、2.5、3h,用pH计检测细胞培养液pH值变化;用MTT法检测第1、2、3、4、5、6、7天细胞增殖情况。结果处理结束后CO2气腹组细胞培养液pH值呈弱酸性,与对照组比下降非常显著(P<0.01),处理后2hpH值恢复正常;He气腹组pH值呈弱碱性,与对照组比升高显著(P<0.05),1.5h恢复正常。MTT法检测细胞增殖结果显示,CO2气腹组和He气腹组人胃癌MKN-45细胞的增殖与对照组比无统计学差异(P>0.05)。结论体外模拟12mmHgCO2气腹和He气腹持续作用3h时,均不促进胃癌细胞增殖。 Objective To study the effects of carbon dioxide or helium pneumoperitoneum on the human gastric cancer cells in terms of proliferation and to provide evidence for the safety of CO2 pneumoperitoneum in laparoscopic radical distal gastrectomy. Methods The cultured gastric cancer cell line MKN-45 was exposed to CO2 or helium environment maintained at 12 mmHg for 3 h. The cells exposed to 5% CO2 at 37 ℃ served as control. In every half an hour during 3-hour exposure, the pH value of culture media was measured. On each day in the following week after exposure, the proliferation of MKN-45 was examined by MTr method. Results The pH value of culture media was a little bit lower than 7 after exposed to CO2 environment, significantly different with that of control group (P 〈0.01 ), but it recovered to normal levels :2 h later. The pH value of helium group was a little bit higher than 7, significantly different with that of control group ( P 〈 0.05 ), then down to normal level in 1.5 h. MTF results showed the proliferation of MKN-45 cells showed no significant difference among CO2 group, helium group and control group (P 〉 0.05). Conclusion Simulated CO2 or He pneumoperitoneum at 12 mmHg does not affect the proliferation of gastric cancer cells MKN-45 in vitro.
