摘要
采用反相高效液相色谱法(RP-HPLC)分离测定了食品中的叶黄素。样品制备采用皂化-萃取法提取食品中的叶黄素,通过AgilentHC-C18柱分离,以甲醇-乙腈(体积比为50∶50)为流动相洗脱,流速1.0mL/min,用二极管阵列检测器(PDA)在446nm波长下检测。测定结果显示,在0.830~41.6mg/L范围内,标准曲线呈良好的线性关系(r=0.99997),加样回收率为97.9%~102%,方法精密度RSD(n=5)为1.1%。该方法具有操作简便,结果准确等优点。
The contents of lutein in food are determined by RP-HPLC in this paper. They are extracted by organic reagent after saponification, and detected at 476 nm after separated by Agilent HC-C18 column. The mobile phase is methanol-acetonitrUe (50-50, v/v), and flow rate is 1.0 mL/min. The linear range is 0.83-41.6 mg/L with correctional coefficient 0.9999. The recoveries is between 97.9%-102%, and RSD(n=5) is 1.1%. This method issimple, accurate.
