After hypo osmotic shock and percoll discontinuous density gradient centrifugation, mass active sperm cells were isolated from the fresh pollen grains of Zea mays .The crude plasma membrane proteins were extracted by the method of aqueous polymer two phase protion.In order to further purify the crude PM protein extracts,three different plant lectins(WGA,ConA and RCAⅠ)were coupled to the activated Sepharose 4B separately to three kinds of affinity gel columns(AC).The retained glycoproteins were eluted and enriched by ultrafiltration,and then isolated and identified by SDS PAGE.Through silver staining analysis,there were 7 protein constituents(62,58,37,34,22,18,15 kD)to bind specifically to WGA,6 constituents(68,66,60,39,37,34 kD)to ConA,and 1 constituent(67 kD)to RCA Ⅰ.The result indicated the existence of the glcNAc,mannosyl,and galactosyl residues in the plasma memberane glycoproeins of maize sperm cells.
